[Interleukin-33/ST2: a new signaling pathway in immunity and immunopathology].

Toll like receptors (TLR) čine familiju transmembranskih signalnih proteina koji delujući kao senzori za produkte mikroorganizama započinju antimikrobni imunski odgovor. Do danas, kod sisara je poznato 12 TLR gena koji kodiraju isto toliko membranskih glikoproteina sa ekstracelularnim delom bogatim leucinom i intercitoplazmatskim delom sa homologijom (TIR). Toll-IL-1 receptor (TIR) domen sadrži konzerviranu proteinsku sekvencu koja je, osim u ovim receptorima, prisutna i u citoplazmatskom delu receptor tipa I za interleukin-1 (IL-1R1). Citokini koji pripadaju familiji IL1 ostvaruju svoju funkciju vezujući se za istoimenu familiju receptora (IL-1R), koji pripadaju superfamiliji Toll-like/IL-1 (TLR-IL-1R) receptora. ST2 gen, prvobitno identifikovan je na mišjim fibroblastima (poznat i kao T1, DER 4, Fit 1) kodira ST2 protein koji je član familije IL-1R . Kao i svi ostali geni IL-1R podgrupe i ST2 se nalazi na mišjem hromozomu 1 4 i humanom hromozomu 2 . Mišji ST2 gen kodira dve mRNA: kraću za sekretornu formu ST2 (sST2) i dužu za transmembransku formu ST2 (ST2L) molekula. Humani ST2 gen uz ove kodira još dve mRNA i to za varijante ST2V i ST2LV . Solubilni ST2 je identičan sa ekstracelularnim regionom ST2L, osim u dodatnih devet aminokiselina koje predstavljaju C-terminus sST2 7 molekula. Solubilnu formu ST2 molekula stvaraju mnoge ćelije u organizmu: fibroblasti, makrofagi, limfociti Th2, ćelije karcinoma dojke, mnoge embrionalne ćelije 5, , dok je membranska forma eksprimirana na mastocitima, bazofilnim i eozinofilnim granulocitima, dendritskim ćelijama, makrofagima, Th2 limfocitima, NK i NKT 1, 9, 11 ćelijama (slika 1).
